Can I use this device to backup both my PC and an external drive connected to my PC via USB (e.g., My Passport)?

I don't see why not.

I'm currently doing this with my Mac. I have it set to do a Thursday backup to one folder and use it as an external drive in other folders. The thing I cannot do with this method is to have a bootable backup. With that said, I could partition the drive to make my computer view it as two harddrives. I could then make one of the two bootable.

So in answer to your question with the PC. Yes. Of course. Just make a subfolder on the drive and setup your automatic backup settings to store the backup in a folder inside the root of the drive. With that said, this is true with any harddrive, not just the model.
